Abstract

The authors studied the effects of estrogen deficiency and hormone replacement therapy on the biomechanical properties of small arteries in rats in vitro. 20 nulliparous, non-pregnant female Sprague-Dawley rats were pharmacologically overiectomised by 750 μg/kg triptorelin im. every 4th week and received either 450 μg/kg estradiol propionate or its vehicle im. every week. Ten other animals received only the vehicles of triptorelin and estradiol (control group). After 12 weeks of treatment cylindrical segments of the saphenous artery were isolated and cannulated at both ends. Pressure- diameter curves in the range of 0-200 mmHg were recorded from segments in normal KR with norepinephrine (1.6x10-5 M) and than with papaverine (2.8x10-5 M). The vessel segment near to the examined one was sent for morphological examination. Biomechanical parameters of the vessel wall were calculated based on the pressure diameter curves. Pharmacological ovariectomy decreased, estrogen replacement increased significantly the passive diameter of the arteries. Intrinsic myogenic tone was decreased after castration and restored by estrogen replacement. Reactivity of the vessel wall to norepinephrine was decreased by estrogen deficiency, hormone replacement therapy induced significantly increased vasoconstriction. No histological changes were seen in the examined vessels. Our results are the first description on hormone replacement effects on the biomechanical properties of small arteries. We have found a new, peripheral mechanism for the development of perimenopausal hot flushes as well.
