A simple and widely applicable technique has been elaborated for enrichment of auxotrophic and thermosensitive mutants of Schizosaccharomyces pombe. The method is based on the selective action of cell-wall lytic enzymes from the digestive juice of Helix pomatia and of enzymes from Trichoderma viride. Under conditions where wild-type cells in mixed cultures of old wild-type and mutant cells are selectively induced to grow, the wild-type cells can be lysed and eliminated. The efficiency of the method was increased by using 2-deoxyglucose, an inhibitor of cell-wall sythesis. Cells of Schizossacharomyces pombe, growing in a medium containing glucose and 2-deoxyglucose at a ratio of 40 : 1, were highly sensitive to the lytic effects of enzymes from Helix pomatia digestive juice and from Trichoderma viride.
