A resonance raman study of carotenoid susceptibility to photooxidatiom in the cyanobacterium Anacystis nidulans

Abstract

Carotenoids were proven to be convenient Raman active probes of membrane structure. When no change is detectable in relative peak intensities and positions in the Raman spectrum the sensitivity of carotenoids to photodestruction may offer a new dimension in membrane research. This was taken advantage of by studying phycocyanin-carotenoid interaction in the cyanobacterium Anacystis nidulans. During nitrate starvation the auxiliary pigment phycocyanin is lost and the value of the decay constant calculated from the deterioration of the resonance Raman spectra also diminished. The temperature dependence of the decay constant made possible the determination of the phase transition temperature of the thylakoid membrane as well.
