The evaluation of electron microscopic cytochrome-oxidase reaction in experimental heart muscle hypoxia

Abstract

The effects of acute carbon monoxide hypoxia on the mitochondrium was studied in isolated Langendorff hearts, perfused under either aerobic or histotoxic hypoxic conditions, with or without a dialysis concentrate from deproteinized calf blood 'Solcoseryl(R)'. The anaerobic Krebs-Henseleit solution contains 45.1% CO+4.7% CO2+50.2 N2. After 30 min of exposure, a damage of the mitochondria and a decrease in the cytochrome-c-oxidase enzyme activity were evident. The modified diaminobenzidine (DAB) method was suitable for electron microscopical demonstration of the enzyme activity changes under pathological conditions. The new data suggest that any degeneration of myocardial cells as a result of histotoxic CO-injury is associated with degenerative changes of the mitochondria. The blood dialysate reduced the amount of ultrastructural and functional damage.
