The blood perfusing the adrenal glands in rats anesthetized with thiopental was collected. Steroids were extracted by methylene chloride. The dry residue was placed on Silufol plates and chromatograms were developed in a chloroform methyl alcohol water (95:5:0.4) system. 18OH DHO was eluted by methyl alcohol and its quantity was determined spectrophotometrically at 240 nm. The hormone was identified by means of rechromatography, by use of the Porter Silber reaction and by gas liquid chromatography. The rate of secretion of 18OH DHO was 11.4 ± 0.71 mcg/hr. Hypophysectomy depressed 18OH DHO secretion.
