Methods are described for the efficient separation of ascorbigen, 1′-methylascorbigen, and other possible molecules with different substituants on the indole and ascorbic acid skeleton by normal and reversed phase thin layer chromatography. The UV spectra of ascorbigen and 1′-methylascorbigen obtained by spectrophotometric measurement were compared with those obtained by in situ spectroscopy on the sorbent layer. The detection limits also were determined for ascorbigen and 1′-methylascorbigen using different methods of visualization and densitometric evaluation of the spots or bands.
