Raman study of isolated and "in situ" T7 phage DNA: conformation and possible interaction with the proteins

Abstract

The Raman spectrum of T7 phage is shown to be dominated by the vibrations of the constituent DNA (backbone chain and bases). These vibrations are compared with their counterparts in the spectrum of isolated T7 DNA. Inspite of the great similarity between the two spectra there are some differences which suggest that the bases play an important role in the DNA-protein interaction. The sites of these interactions appear to involve the CH3 and C=O groups of thymine, C=O groups of cytosine and N7-C8 group of guanine. Some preliminary observations are given on the UV induced changes in isolated DNA.
