Racemization analyses of peptides and amino acid derivatives by chromatography with pre-column derivatization

Abstract

Reversed phase-HPLC on ODS-Hypersil column with methanol-acetonitrile-acetate buffer (pH 4) and precolumn derivatization with Marfey's reagent were used to monitor racemization in amino acids and peptides and their protected derivatives by separation of stereoisomeric amino acids. The rate of racemization was measured in bologically active peptides including thymopoietin and FMDV-fragments, in branched peptides based on polylysine, in cyclo- and normal endothiopeptides, and in N- and C-protected amino acid derivatives. Chromatographic samples were obtained by acidic and enzymatic hydrolysis of the different starting substances.
