Quantitative cytochemical investigations on Ehrlich ascites tumour cells treated with Vincristine - I. Feulgen cytophotometric investigations

Abstract

Changes in the DNA content of Ehrlich ascites tumour cells in mice treated with 2 mg/kg Oncovin (Lilly) and 2 mg/kg Vincristine (Richter) ssVCR respectively were studied by Feulgen Cytophotometric method during a 25-day period following treatment. Essential changes in the distribution of the DNA content of cells could not be observed on the first day following treatment, the tumour cells could get undisturbed in the various phases of the cycle. From the 2nd day on more and more polyploid cells appeared and on the 3rd and following days cells in the Gl phase disappeared practically completely from the tumour on account of the extensive polyploidization. Between the 8th and 25th days cells containing DNA quantities exceeding the octoploid level diminished considerably and as compared to the control a double preand postsynthetic DNA containing cell population developed. On the 18th day and 25th day Gl phase cells reappeared in the Oncovin treated tumours. Apart from this the two preparations proved to have the same effect.
