Problems in the measurement of macromolecular synthesis using labelled precursors. Determination of the UTP pool in tonsillar lymphocytes

Abstract

The correlation between the 3H uridine incorporation and the rate of RNA synthesis was investigated in human tonsillar lymphocytes. As shown by our experiments, the data of the incorporation have to be corrected for the specific radioactivity of the UTP pool. The UTP pool of lymphocytes was determined by using E. coli RNA polymerase. From the obtained pool size of UTP, and from the ratio of the 3H uridine incorporation and the 3H uridine uptake into the acid soluble fraction of cells, the apparent rate of RNA synthesis was calculated. This value seems to be the true index of the rate of a process in all cases, if the pool size of the precursor is much higher than the amounts of the precursor taken up by the cells.
