Methyl-β-cyclodextrin/paclitaxel aqueous solutions: A tool to test Cremophor-free paclitaxel

Abstract

Aqueous ethanol paclitaxel solutions with 4.0 mg/ml dissolved paclitaxel concentration can be prepared in 96% ethanol/water in a 2/1 mixture in the presence of 200 mg/ml methylated-β-cyclodextrin. No drug precipitation from the pre-concentrate formulation was observed after more than six months of storage at room temperature exposed to light. No chemical degradation of paclitaxel in this pre-concentrate solution during a six-month storage was detected. The paclitaxel solutions diluted to a clinically feasible concentration were found to remain clear for 8 h after dilution. The paclitaxel/methylated-β-cyclodextrin solutions exhibit appropriate cytotoxicity on number of human cancer cell lines, providing the possibility to test the antitumour activity of paclitaxel without Cremophor as a vehicle.
