The significance of genetic factors in leukemia was examined. In the 1,588 relatives of 120 children with leukemia the incidence of leukemia and malignant tumors was significantly higher than in the general population (p<0.01). After the influence of a chemical mutagen in in vitro experiments the number of chromosomal aberrations in leukocyte cultures was found to be increased in a proportion of the leukemic patients over the findings in normal subjects. On the basis of these findings, the possibility of a genetic predisposition or increased disposition to mutation as an etiological factor in leukemia and malignant tumors is discussed.
