In our work a SECM constructed in our laboratory was used to image the concentration profiles of different species inside the reaction layer of specially prepared biosensors during their operation. The effect of different working parameters on the concentration profiles was investigated. The measured data were compared with the values obtained by digital simulation. In most of the cases the measured dependencies were in agreement with the expectations. We were able to estimate the optimal thickness of the reaction layer - in this case it was about 200 μm. The results proved that the SECM measurements can give important experimental data to prepare enzyme sensors of improved measuring properties.
