Influence of the cultivation conditions in bioreactor on the phenotypic properties of E. coli leu X- strain

Abstract

Geneticaly manipulated E. coli leu X- strain was cultivated in a bioreactor in comparison with a wild type strain to develop a method to receive big quantity of bacterial cells. The influence of different cultivation conditions on the phenotypic markers were studied. The best growth of the mutant strain was observed at automatic regulation of pH 7,2, leval of dissolved oxygen 10 or 50% and glucose addition dropwise during the cultivation in quantities 20 or 10g/l in Luria broth enriched with trypticase peptone. No changes in the phenotypic features were observed at the studied cul tivation conditions. There were no changes in the virulence to mice after intraperitoneal challenge with the wild or mutant strains cultivated static or in bioreactor. Vaccine from the leu X- strain protected mice against intraperitoneal challenge with the wild strain.
