Flowzytometrische Leukozytenzählung in Thrombozytenkonzentraten

The flow cytometer “CytoronAbsolute” was used for counting of leukocytes in leukocytepoor platelet concentrates. The leukocytecounts were determinedin platelet concentrates firstbyconveritionalmethods (Sysmex K1000 counter, Bürker and Nageotte Chambers) and then by flow cytometer. Leukocytes were counted in a range of 0.08 - 5.500/μl and fluorescence events were detected by the flow cytometer. The linearity between expected counts and events found by the flow cytometer was excellent above a leukocyte concentration of 500/μl. In the range of 0 - 500/μl, however, a double logarithmic approach resulted in a stronger correlation between expected and found leukocyte counts. The leukocyte concentration in random-donor leukocyte-poor platelet concentrates was determined as 30.9 ± 27.1/μl (mean ± SD, n = 97) and that of apheresis products as 6.2 ± 6.9/μl (n = 100).
