For the demonstration of aryl sulfatase in the corpus pineale, p-nitrocatechol sulfate (2-hydroxy-5-nitrophenyl-sulfate) was used as a substrate. The reaction is negative when p-nitrophenylsulfate or acetylphenylsulfate are used. Localization of the enzyme was seen in the Golgi system and in the smooth surfaced endoplasmic reticulum. By this histochemical reaction the formation of primary lysosomes can be demonstrated on the level of the Golgi system and of the smooth-surfaced endoplasmic reticulum as well. Arylsulfatase corresponds to the A and B type. The reaction can be inhibited by the presence of parachloromercuribenzoate and KCN.
