Abstract

The authors investigated the effects of medroxyprogesterone monotherapy and combined hormone replacement on the biomechanical properties of small arteries in rats in vitro. 30 nulliparous, non-pregnant female Sprague- Dawley rats were pharmacologically castrated with 750 μg/kg triptorelin im. every 4th week and received either 15 mg/kg medroxyprogesterone acetate im. biweekly or the above-mentioned treatment plus 450 μg/kg estradiol propionate im. once a week, or vehicles of these steroids (10 animals in each group). 10 other animals (control group) received only the vehicles for the same period of time. After 12 weeks of treatment cylindrical segments of the saphenous artery were isolated and cannulated at both ends. Pressure-diameter curves were recorded from segments in normal Krebs-Ringer (KR), with norepinephrine (1.6x10-5M), and then with papaverine (2.8x10-5M) in the range of 0-200 mmHg. Biomechanical parameters of the vessel wall were calculated based on the pressure-diameter curves and the wet weight of the specimen. The results were compared to those of their previous study, in which oestrogen monotherapy had been investigated. Chemical oophorectomy decreased the passive diameter of the arteries medroxyprogesterone monotherapy did not alter the vessel diameter, its effect being the same as that of estradiol monotherapy. The reactivity to norepinephrine and papaverine was not changed significantly neither medroxyprogesterone monotherapy nor after the combined hormonal replacement. These results are the first description of the changes in the biomechanical properties of small arteries after progestagen monotherapy or combined hormonal replacement therapy.
