Cytochemistry of tubuloreticular structures in lymphocytes from patients with systemic lupus erythematosus and in cultured human lymphoid cells. Comparison to a paramyxovirus

Abstract

Tubuloreticular structures (TRS) which occur in the blood lymphocytes of patients with autoimmune disease and which can be induced in human lymphoid cell cultures by treatment with 5 bromodeoxyrudine were investigated cytochemically. In samples fixed with glutaraldehyde or osmium tetroxide and embedded in epoxy resin, the TRS appeared as networks of 20 to 30 nm. diameter membranous tubules within the endoplasmic reticulum. An electron dense material associated with the tubules was highlighted after embedding in glycol methacrylate or after fixation with potassium permanganate. The glycol methacrylate embedded material resisted digestion by pancreatic ribonuclease or deoxyribonuclease and stained intensely with the Rambourg method for glycoprotein. Although the constituent tubules of TRS are of 'virus like' proportions, there was no evidence for a relationship to similarly sized cores of a known paramyxovirus. In glycol methacrylate sections of chicken embryo cells infected by Newcastle disease virus, cytoplasmic inclusions of the paramyxovirus nucleocapsids were digested by the pancreatic ribonuclease treatment. The findings suggested that TRS consist principally of phospholipid and acidic glycoprotein.
