For the isolation of the platelet plasma and the cytoplasmatic membranes, the following methods were applied: glycerol-osmotic lysis, combined with sonication; glycerol-osmotic lysis carried out on fresh or frozen platelets; and detergent treatment followed by separation of the protein fractions insoluble at low ionic strength. The detergents used were Triton X-100, Nonidet P-40, Na-deoxycholate and digitonin. The protein fractions were compared by sodium dodecyl sulfate (SDS)-polyacrylamide gel electrophoresis (PAGE).
