Changes of Plasma Membrane H+-ATPase in Pathogen-resistant and Pathogen-sensitive Near-isogenic Wheat Lines in Response to Elicitor from Stem Rust

Abstract

The aim was to investigate the effect of elicitor prepared from wheat stem rust on H+-ATPase activity in plasma membranes of near-isogenic wheat lines SrHopeRa and SrHopeSa, resistant and susceptible to stem rust race 1 pathogen, respectively. Plasmalemma fraction was purified from crude microsomal fraction by means of the phase separation in aqueous two-phase system of dextran T500 and PEG 4000 polymers. Specific H+-ATPase activity in resistant line was significantly higher than from susceptible one. In the presence of small amount of elicitor activity of the enzyme increased significantly only in susceptible line, but it did not reach the level of resistant one. In the resistant line significant increase of H+-ATPase activity was observed only after adding 4-times higher dose of elicitor. The influence of elicitor on the enzyme kinetics was exactly the same both in Ra and Sa membranes, changes of both Km and Vmax values as influenced by elicitor were less than 1%. It may be assumed that the presence of the stem rust resistance gene in Ra line results only in the increase of amount the H+-ATPaSe, however not as appearance of new molecular form of the enzyme.
