Application of TLC - Digital autoradiography as a rapid method in a pilot study of deramciclane metabolism

Abstract

A pilot study has been conducted to develop sample preparation and semi-quantitative determination of metabolites formed from deramciclane (a new anxiolytic compound) in experimental animals. A solid phase extraction procedure was developed for isolation of radiolabeled compounds from excreta. Eluents for TLC separation of the isolated compounds were optimized. The study has shown that digital autoradiographic evaluation of thin layer chromatograms is a valuable tool for obtaining rapid information; it also enables the analyst to perform in situ semi-quantitative analysis of compounds separated. Several applications are given.
